Polyclonal Antibody Sequencing.

Use REpAb® polyclonal antibody sequencing to convert your pAb to a set of mAbs. Get the robust performance of your polyclonal antibody and the reliability of recombinant antibodies, by sequencing the antibodies within the pAb and recombinantly expressing the dominant form(s).

How Polyclonal Antibody Sequencing Works.

Through proprietary chemistry, separation, sample preparation and artificial intelligence, dozens of mass spectrometry experiments come together to derive the full sequences of mAbs within a polyclonal mixture. The workflow involves multiple enzymes and uses bottom-up, top-down and middle-down mass spectrometry. AI-powered de novo sequencing and assembly algorithms are used to overlay the enormous data set to determine the full antibody sequences.

REpAb is the only commercial polyclonal antibody de novo protein sequencing platform – we are uniquely capable of sequencing antibodies from a complex polyclonal mixture without reference to a sequence database or genomic information.

Applications of Polyclonal Antibody Sequencing.

As a stand-alone service, polyclonal antibody sequencing is most commonly used in development of in-vitro diagnostics (IVD). IVD that are based on pAbs are highly dependent on the producing animal, and are subject to costly revalidation and batch-to-batch variability. Converting them to rmAbs leads reduces supply chain risk, and eliminates revalidation concerns.

Getting Started With Polyclonal Antibody Sequencing.

Requirements

  • 1 mg polyclonal antibody – must be antigen-affinity and protein A/G purified IgG
  • Most species (rabbit, chicken, goat, sheep, camelid, dog, etc.)
  • Single host (not pooled); pooling from multiple bleeds of a single host is acceptable
  • Optional: 100 μg antigen protein
  • Optional: NGS data or PBMCs from the same host at a similar time point
