Beyond binding assays: specificity plus reproducibility.

Since the protein sequence determines structure and function, rapid confirmation of the protein sequence offers an ideal complement to performance validation assays when revalidating antibodies. Validation is important: recent studies have shown that at least 50% of reagent antibodies cannot recognize their targets specifically, and half or more preclinical studies cannot be reproduced. But validation can be cumbersome especially as multiple complementary validation experiments should repeated on each new batch. Furthermore, properties other than binding behavior are often overlooked – causing problems downstream. A much more precise, repeatable, and full confirmation of the protein sample ensures the antibody is as expected, reducing the need for revalidation or troubleshooting downstream.

Getting Started is Easy.

Here’s what we need:

  1. 20 μg of each protein of interest
  2. Reference sequence(s) in FASTA
  3. The sample should be reasonably pure and not a polyclonal mixture.

You get:

  1. Confirmation that the protein of interest is in your sample
  2. Coverage down to the amino acid level
  3. Any other observations for further investigation (e.g. suspected mutation, PTM, other dominant forms)

Don’t have the sequence yet?
