Antibody Fc Receptor Profiling.

Predict Effector Function and Pharmacokinetics with High-Precision Biacore SPR Analysis.

What is Fc Receptor Profiling?

Fc receptor profiling involves the quantitative measurement of the interaction between the Fc (fragment crystallizable) region of an antibody and various cell surface receptors. These interactions are the primary drivers of an antibody’s biological activity beyond antigen binding. By characterizing the binding kinetics (kon, koff) and affinity (KD), researchers can determine how effectively an antibody will trigger immune responses or how long it will remain circulating in the body.

Why is Fc Receptor Profiling Essential for Antibody Development?

Characterizing the Fc region is a regulatory and functional requirement for biotherapeutic development. It allows for the fine-tuning of an antibody’s profile to match its intended therapeutic use:

  • Effector function prediction
  • Pharmacokinetics and Half-Life
  • Fc Silencing and Effector Function Knockout
  • Safety and Efficacy

How Fc Receptor Profiling works.

Rapid Novor provides comprehensive Fc receptor (FcR) profiling through our RapidSPR™ platform, utilizing the industry-leading Biacore 1K system and our comprehensive Fc receptor panels.

Our specialized binding assays for Fc gamma receptors (FcγRs) and neonatal Fc receptor (FcRn) offer critical insights into the therapeutic potential of your antibody candidates, enabling the prediction of effector functions and serum half-life early in the development cycle.

We utilize the Biacore 1K SPR system to perform high-sensitivity, label-free analysis of these interactions, ensuring that even low-affinity binding events are characterized with precision.

RapidSPRTM Comprehensive Receptor Panels.

Our RapidSPR™ service covers the full spectrum of relevant Fc receptors. We offer broad panel screens, or more focused cohorts if you just have a couple of candidates. We can provide panels, or adapt to your custom needs.

Below are typical targets featured within our panel offerings. While these represent common setups, our platform accommodates any custom target sequence required for your project. We can work with our trusted vendors on your behalf to order fresh panels at the start of each project.

Receptor Category Specific Receptors Predicted Biological Function
High-Affinity FcγR CD64 (FcγRI) Potent trigger of ADCP and pro-inflammatory cytokine release.
Low-Affinity FcγRs CD16 (FcγRIIIa/b), CD32 (FcγRIIa/b/c) Drivers of ADCC (CD16) and inhibitory/activatory immune signaling.
Salvage Receptor FcRn Regulation of IgG and albumin homeostasis; determines serum half-life.

RapidSPRTM Biacore Platform.

Surface Plasmon Resonance (SPR) is the “gold standard” for biomolecular characterization. While other methods like ELISA or Bio-Layer Interferometry (BLI) provide endpoint data, Biacore SPR offers clear advantages:

The Rapid Novor Difference:

Complex Half-Life Analysis.

All FcRn projects come with custom half-life analysis.

  • Our custom analysis measures the exact time required for half the antibody-FcRn complex to dissociate (t1/2), giving a clear readout of residence time at pH 6.0 versus at pH 7.4.
  • Complex half-life is a better predictor of antibody clearance in vivo than a standard affinity ranking, allowing you to confidently identify and advance top candidates.
